Clinical Experience of Mioreol® Use in the Treatment of Dementia Caused by Alzheimer’s Disease
Abstract
The relevance of the study is the urgent need to find new treatment options for Alzheimer’s disease (AD) to reduce cognitive deficits and neuropsychiatric symptoms. The aim of this study was to analyze the efficacy and safety of Mioreol® in patients with moderate to moderate-severe dementia due to AD in a non-interventional observational programme.
Materials and methods. The study was conducted in 30 patients with moderate and moderate-severe AD aged from 61 to 90 years (median age 73.5 [68.00; 78.00]). Nineteen patients had moderate dementia and 11 had moderate — severe dementia. The therapeutic dose of Mioreol® was 10 mg donepezil + 20 mg memantine, taken orally once a day at the same time, independent of meals. The duration of study was 24 weeks. The effects of the drug were assessed using the MMSE, ADAS-Cog, NPI and CGI scales at baseline and by the end of 12 and 24 weeks of treatment.
Results. The use of Mioreol® in a six-month therapy of AD patients with moderate and moderate-severe dementia showed a positive effect (CGI-C scale) in 54.2 % of the total number of AD patients; to note, an improvement was observed in 80 % of patients with moderate stage of dementia caused by AD. During the Mioreol® therapy cognitive function, psychotic and behavioral symptoms significantly improved. In majority of patients with moderate-severe AD, cognitive was observed in, severity of behavioral and psychotic symptoms did not change significantly compared to baseline.
Conclusion. Fixed-dose combination therapy with Mioreol® (donepezil10 mg/memantine 20 mg) is effective and well tolerated in patients with moderate and moderate-severe AD. The use in patients with moderate stage of dementia caused by AD is more effective. The use of Mioreol® with fixed doses of doneperil and memantine is more convenient than taking two separate drugs twice a day, which improves adhence to therapy for these patients.
